Thymosin beta-4 was isolated from calf thymus in the early 1980s and later characterised as an abundant intracellular actin-sequestering protein. Interest in short synthetic fragments grew once the actin-binding motif had been mapped to the middle of the sequence. TB-500 came out of that line of work as a truncated analogue rather than a natural isolate, and it is now sold mainly to laboratories. Published studies on the fragment have been largely in vitro or in animal models, and controlled human trials remain sparse, so claims about effects in people rest on extrapolation.
Literature and online discussion often conflate TB-500 with full-length thymosin beta-4, even though the two differ in size and are not interchangeable in analytical terms. The fragment is produced by solid-phase peptide synthesis, and the product is a defined seven-residue chain rather than a biological extract. Because the term is a trade-style label, two vendors may supply materials of the same nominal sequence but different counter-ion content, purity, or water content. Comparisons across studies are therefore difficult unless the exact sequence and purity are reported.
Identity and purity checks for peptide material typically combine reversed-phase high-performance liquid chromatography with mass measurement, since retention time alone cannot confirm a sequence. Mass measurement verifies the expected molecular mass within instrument tolerance, while chromatographic peak area provides a purity estimate. Anti-doping analysis of urine uses related but more sensitive workflows, sometimes after solid-phase extraction. For research material, batch documentation, certificate content, and independent testing are common points of scrutiny, because supply chains outside pharmaceutical regulation vary widely in the paperwork they provide.
Reconstitution of a lyophilized peptide is normally done with sterile water or a suitable buffer under aseptic conditions. Adding solvent down the vial wall and allowing gentle dissolution instead of vigorous vortexing reduces the chance of aggregation, which can lower the effective concentration of the resulting solution. Concentrated stocks are usually diluted into working buffer shortly before use. Because no standard preparation protocol exists for TB-500 specifically, laboratories adapt general peptide handling practice, and reported results may reflect differing preparation choices.
Dry peptide powder is commonly kept at −20 °C in a desiccated container away from light, a practice that limits moisture uptake and oxidation. Once dissolved, solutions are generally held at 2–8 °C for short periods or frozen at −20 °C or lower for longer storage, with repeated freeze-thaw cycles avoided. Hydrolysis and oxidation are the main degradation routes for peptides in solution, and both accelerate at higher temperature or extreme pH. Published stability data specific to TB-500 are limited, so shelf life should be treated as uncertain.

Once dissolved, the peptide is far less stable than the dry powder. Aqueous solutions are subject to hydrolysis, oxidation at susceptible residues and gradual loss of material through adsorption onto glass and plastic surfaces. Terminal glutamine can cyclise under some conditions, producing a related species that complicates purity assessment. Dilute solutions tend to lose a larger fraction of material to surfaces than concentrated ones. Buffers, pH and ionic strength all influence the rate of change, so stability figures are only meaningful when those parameters are stated alongside the storage interval.
Detection in biological samples relies on mass spectrometry, typically liquid chromatography coupled to tandem mass spectrometry after peptide extraction and enrichment. Intact peptides can also be confirmed by high-resolution mass measurement together with fragmentation data. Detection windows in urine are short because the peptide is degraded by proteases and cleared quickly, and concentrations are low. Many jurisdictions treat the compound as a prohibited substance in sport, grouped with peptide hormones and related factors, while it is not an approved therapeutic product. Identity and purity statements therefore rest on certificates of analysis, ideally issued by an independent laboratory.
Identity and purity are usually assessed by reversed-phase high-performance liquid chromatography together with mass spectrometry. The chromatogram provides a purity estimate as a percentage of total peak area, while the mass spectrum confirms that the observed mass matches the expected value. Amino acid analysis or tandem mass spectrometry sequencing can provide additional confirmation. Reported purity figures depend on the column, gradient, and detection wavelength, so values from different laboratories are not directly comparable without method details.
Lyophilised peptide is normally reconstituted with sterile water or a neutral buffer shortly before use. Because repeated freeze-thaw cycles can degrade the material, dividing a reconstituted solution into single-use aliquots is a common practice. Working solutions are usually kept cold and protected from light. The exact shelf life depends on concentration, buffer composition, and handling, so it is often determined empirically rather than assumed.
A myristic acid molecule is attached to this analogue, which associates the insulin molecule to the abundant serum albumin, which in turn extends the effect and reduces the risk of hypoglycemia. Both protracted analogues need to be taken only once daily, and are used for type 1 diabetics as the basal insulin. A combination of a rapid acting and a protracted insulin is also available, making it more likely for patients to achieve an insulin profile that mimics that of the body's own insulin release. Insulin is also used in many cell lines, such as CHO-s, HEK 293 or Sf9, for the manufacturing of monoclonal antibodies, virus vaccines, and gene therapy products. Insulin is usually taken as subcutaneous injections by single-use syringes with needles, via an insulin pump, or by repeated-use insulin pens with disposable needles. Inhaled insulin is also available in the U.S. market. Those who take insulin may choose a particular method of administration for improved blood glucose control, discrete delivery, comfort, or cost, among a variety of other factors. Unlike many medicines, insulin cannot be taken by mouth because, like nearly all other proteins introduced into the gastrointestinal tract, it is reduced to fragments, whereupon all activity is lost. There has been some research into ways to protect insulin from the digestive tract, so that it can be administered orally or sublingually. In 2021, the World Health Organization added insulin to its model list of essential medicines.
=== Insufflation === Another popular route to intake heroin is insufflation (snorting), where a user crushes the heroin into a fine powder and then gently inhales it (sometimes with a straw or a rolled-up banknote, as with cocaine) into the nose, where heroin is absorbed through the soft tissue in the mucous membrane of the sinus cavity and straight into the bloodstream. This method of administration redirects first-pass metabolism, with a quicker onset and higher bioavailability than oral administration, though the duration of action is shortened. This method is sometimes preferred by users who do not want to prepare and administer heroin for injection or smoking but still want to experience a fast onset. Snorting heroin becomes an often unwanted route once a user begins to inject the drug; a user may still get high on the drug from snorting and experience a nod, but will not get a rush because the drug is absorbed slowly rather than instantly.

=== Detection === Modafinil is considered a stimulant doping agent and as such is prohibited by World Anti-Doping Agency in sports competitions. Modafinil enantiomers can be separately quantified in biological samples.
This is based on the absence of Neanderthal mtDNA, as well as the relatively low level of Neanderthal admixture in the modern human X chromosome. According to Svante Pääbo, it is not clear that modern humans were socially dominant over Neanderthals, which may explain why the interbreeding occurred primarily between Neanderthal males and modern human females. A 2026 study confirmed the evidence of sex bias, reporting that sexual selection (i.e., a preference for male Neanderthals and modern human females) was the primary driver of the asymmetric mating patterns, rather than sex biases in demography or migration. Neanderthals in the Siberian Altai Mountains interbred with the local Denisovan population, and it may have been a common occurrence here. About 17% of the genome of one Altai Denisovan specimen derived from Neanderthals. Even before genetic evidence had confirmed this admixture, hybridisation between Neanderthals and early modern humans had been proposed early on, such as by English anthropologist Thomas Huxley in 1890, Danish ethnographer Hans Peder Steensby in 1907, and Coon in 1962. In the early 2000s, supposed hybrid specimens were discovered: Lagar Velho 1 and Muierii 1. These proposals were based on shared anatomy, but modern geneticists have cautioned that some shared anatomy might have been acquired by adaptation to a similar environment, rather than admixture alone.

In 1972, the restaurant was selected as a feature location by George Lucas for his 1973 film American Graffiti. The Mel's used was located at 140 South Van Ness Avenue in San Francisco. It serves as the setting for the opening scene of the film as well as the backdrop for the opening credits, accompanied on the soundtrack by Bill Haley’s “Rock Around the Clock”. The prominent play given to the location has been credited with having saved the company from possibly going out of business. Signage and artwork from the Mel's chain are frequently used in marketing for the film. Universal Studios has built replica Mel's Drive-In restaurants, resembling the one featured in American Graffiti, at their theme parks in Hollywood, Orlando, Japan, Beijing, and Singapore. Prior to American Graffiti, Mel's was used as a location in the 1967 film Guess Who's Coming to Dinner. Spencer Tracy and Katharine Hepburn are out for a drive, and Tracy pulls into Mel's and orders Oregon boysenberry ice cream; he then has a minor traffic altercation with a black man. The Mel's was located in the Excelsior district of San Francisco. Hepburn and Tracy never actually visited the location. Mel's restaurants have since been featured in other media, such as Melrose Place (1996, Season 5, Episode 1), Doonesbury comics (December 18, 1989), and the book The American Drive-in by Mike Witzel.
== Therapeutics == Several mimetics of ELA have been developed for therapeutic purposes. Amgen has created a camel antibody and a small molecule agonist capable of mimicking the function of ELA towards it cognate receptor APLNR. The latter has entered phase 1 clinical trials for heart failure and acute kidney disease. Bristol Myers Squibb has also created its own small molecule agonist of APLNR. An opinion published in the Lancet in 2019 suggested that ELABELA could be used to treat intrauterine growth restriction and maternal morbidity linked to eclampsia.
The small intestine starts at the pyloric sphincter, and finishes at the ileocecal valve. Partially digested food starts to arrive in the small intestine as semi-liquid chyme, one hour after it is eaten. The stomach is half empty after an average of 1.2 hours. After four or five hours the stomach has emptied. In the small intestine, the pH becomes crucial; it needs to be finely balanced in order to activate digestive enzymes. The chyme is very acidic, with a low pH, having been released from the stomach and needs to be made much more alkaline. This is achieved in the duodenum by the addition of bile from the gall bladder combined with the bicarbonate secretions from the pancreatic duct and also from secretions of bicarbonate-rich mucus from duodenal glands known as Brunner's glands. The chyme arrives in the intestines having been released from the stomach through the opening of the pyloric sphincter. The resulting alkaline fluid mix neutralises the gastric acid which would damage the lining of the intestine. The mucus component lubricates the walls of the intestine.
Of the 240 planes that were launched for the strike, 14 aborted for various reasons and returned to their ships. The 226 planes that continued consisted of 95 Hellcat fighters (some carrying 500-pound bombs), 54 Avenger torpedo bombers (only a few carrying torpedoes, the rest four 500-pound bombs) and 77 dive bombers (51 Helldivers and 26 Dauntlesses). The TF 58 aircraft arrived over the Japanese fleet just before sunset. The 35 or so fighters Ozawa was able to put up were overwhelmed by the 226 incoming aircraft of Mitscher's attack. While the few Japanese aircraft were often skillfully handled and the Japanese anti-aircraft fire was intense, the U.S. planes were able to press in on the attack. The first ships sighted by the U.S. strike were oilers, 30 miles (48 km) before the carrier groups. The strike group from Wasp, more concerned with their low fuel levels than with finding the more important Japanese carriers and battleships, dived on the tankers. Two of these were damaged so severely that they were later scuttled, while a third was able to put out fires and get underway. The carrier Hiyō was attacked and hit by bombs and aerial torpedoes from four Grumman TBF Avengers from Belleau Wood. Hiyō was set afire after a tremendous blast from leaking aviation fuel. Dead in the water, she sank stern first, with the loss of 250 officers and men. The rest of her crew, about one thousand, were rescued by Japanese destroyers. The carriers Zuikaku, Junyō and Chiyoda were damaged by bombs.

On 26 November 2018, the mayor of Moscow, Sergey Sobyanin, participated in the ceremony to launch the cable car above the Moskva River. This car connects the Luzhniki sports complex with Sparrow Hills and Kosygin Street. The trip from the viewpoint on Vorobyovy Gory to Luzhniki Stadium takes only five minutes, rather than 20 minutes by car.
=== Climate change impacts === Increasing sea surface temperatures in tropical regions (~1 °C (1.8 °F)) the last century have caused major coral bleaching, death, and therefore shrinking coral populations. Although coral are able to adapt and acclimate, it is uncertain if this evolutionary process will happen quickly enough to prevent major reduction of their numbers. Climate change causes more frequent and more severe storms that can destroy coral reefs. Annual growth bands in some corals, such as the deep sea bamboo corals (Isididae), may be among the first signs of the effects of ocean acidification on marine life. The growth rings allow geologists to construct year-by-year chronologies, a form of incremental dating, which underlie high-resolution records of past climatic and environmental changes using geochemical techniques. Certain species form communities called microatolls, which are colonies whose top is dead and mostly above the water line, but whose perimeter is mostly submerged and alive. Average tide level limits their height. By analyzing the various growth morphologies, microatolls offer a low-resolution record of sea level change. Fossilized microatolls can also be dated using radiocarbon dating. Such methods can help to reconstruct Holocene sea levels. Though coral have large sexually-reproducing populations, their evolution can be slowed by abundant asexual reproduction. Gene flow is variable among coral species.

=== Detailed expression of Thy-1 === In mice, Thy-1 is also found on thymocytes, peripheral T cells, myoblasts, epidermal cells, and keratinocytes. It is one of the "pan T cell markers"(of mice) like CD2, CD5 and CD28. In humans, Thy-1 is also expressed by endothelial cells, smooth muscle cells, a subset of CD34+ bone marrow cells, and umbilical cord blood-, cardiac fibroblasts, and fetal liver-derived hemopoietic cells. Thy-1 is present on a fraction of brain cells and a fraction of fibroblasts of most vertebrate species studied. Nervous tissue: Thy-1 expression in the nervous system is predominantly neuronal, but some glial cells also express Thy-1 especially at later stages of their differentiation. One study compared Thy-1 expression in four human neuronal cell lines, two neuroglial cell lines, and fresh tumor cells of neuronal origin and found three of the four neuronal cell lines, all of the neuroglial cell lines, and 80% of the tumors to be strongly positive for Thy-1. Brain part specific ELISA reports are available which show highest concentrations of Thy-1 protein in the striatum and hippocampus, followed by the neocortex, cerebellum, spinal cord, and the retina and optic nerve. Thy-1 promoter has often been assumed to be "brain specific". "Neuron specific" mouse Thy-1 promoter has been used to drive "brain specific" forced expression of proteins e.g. mutated Amyloid precursor protein(APP) as transgenic animal models of Alzheimer's disease. Thy-1 expression in the brain is developmentally regulated.
Asparagine peptide lyase are one of the seven groups in which proteases, also termed proteolytic enzymes, peptidases, or proteinases, are classified according to their catalytic residue. The catalytic mechanism of the asparagine peptide lyases involves an asparagine residue acting as nucleophile to perform a nucleophilic elimination reaction, rather than hydrolysis, to catalyse the breaking of a peptide bond. The existence of this seventh catalytic type of proteases, in which the peptide bond cleavage occurs by self-processing instead of hydrolysis, was demonstrated with the discovery of the crystal structure of the self-cleaving precursor of the Tsh autotransporter from E. coli.

No. Thymosin beta-4 is a 43-residue protein, while TB-500 matches only residues 17 to 23 of that chain. The two are related but differ in size, and a method that identifies one does not automatically identify the other.
It corresponds to the actin-binding region of thymosin beta-4. Commercial synthesis reproduces that seven-residue stretch, usually with an acetyl group on the N-terminus.
It is not authorised as a medicine in the United States or the European Union and is distributed as a research chemical. Labels consequently carry research-use-only statements rather than clinical indications.
Standard practice is a desiccated container at −20 °C, protected from light and kept sealed between uses. Letting the vial reach room temperature before opening reduces condensation on the powder. Repeated warming and cooling of the whole container is generally avoided.
